Enzymes are biological catalysts that accelerate specific reactions without being consumed by the reaction. Their selectivity can make them valuable tools for producing complex pharmaceutical intermediates and active ingredients.

What is an enzyme?

An enzyme binds a suitable substrate at its active site and lowers the energy needed for a reaction. Enzyme performance depends on factors including pH, temperature, substrate concentration, product concentration and the surrounding solvent system.

Pharmaceutical applications

Enzymes support selective hydrolysis, acylation, deacylation, chiral resolution and other biotransformations used in antibiotic intermediates, active pharmaceutical ingredients and speciality pharmaceutical products.

Penicillin G amidase

One enzyme, two useful directions.

Penicillin G amidase—also known as penicillin G acylase—hydrolyses the side-chain amide bond of penicillin G to produce 6-APA and phenylacetic acid. Under controlled synthesis conditions, acylase activity can also be used to attach a suitable side chain to a beta-lactam nucleus.

Supporting penicillin and cephalosporin pathways

Acylases remove or transfer acyl groups. In cephalosporin processing, suitable acylase systems can support conversion of acylated intermediates toward 7-ACA. Enzyme choice must match the exact substrate: activity toward one intermediate does not guarantee efficient activity toward another.

Where enzyme performance can be lost

Laboratory activity does not automatically translate into an efficient production process. Reaction conditions, enzyme lifetime and downstream recovery must be considered together.

pH & temperature

Reaction products can shift pH, while excessive temperature or unsuitable pH can reduce enzyme activity and stability.

Inhibition & equilibrium

High substrate or product levels may inhibit activity. In synthesis reactions, competing hydrolysis can reduce the desired conversion.

Mass transfer

With immobilised enzymes, diffusion through beads or supports can limit the rate even when the enzyme itself remains active.

Lifetime & reuse

Mechanical stress, impurities and repeated cycles can reduce activity, making enzyme stability and reuse validation important.

Downstream recovery

Enzyme selection should consider filtration, crystallisation, impurity control and product isolation—not reaction conversion alone.

Why use an enzyme instead of a conventional chemical route?+

Enzymes can offer high reaction and stereochemical selectivity under comparatively mild conditions. The best route depends on overall yield, raw materials, cycle time, downstream recovery, waste treatment and cost—not selectivity alone.

What controls enzyme activity?+

Important variables include pH, temperature, substrate and product concentration, ionic strength, solvent composition, mixing and reaction time. Each enzyme preparation has its own practical operating window.

What is an immobilised enzyme?+

An immobilised enzyme is attached to or contained within a solid support. This can simplify separation and enable reuse, although support chemistry, particle strength and internal diffusion can affect performance.

Can one acylase work for every substrate?+

No. Enzymes are substrate-selective. Activity observed with penicillin G or one cephalosporin intermediate cannot be assumed for a different nucleus, side chain or activated donor without testing.

What should be measured during scale-up?+

Teams commonly monitor conversion, selectivity, pH demand, temperature, mixing, substrate feed, impurity formation, enzyme activity retention, filtration behaviour and product recovery.
